UTR region of LDLR gene and also, two C>G transversion in SMARCA4 gene. All of these heterozygote changes have been reported for the first time. Conclusion: The results indicate different effects of polymorphisms of LDLR, PCSK9 and SMARCA4 genes on coronary artery disease in differen populations. Keyword: Coronary Artery Disease, LDLR, PCSK9, SMARCA4, Polymorphism, Genotype-Phenotype, IranبUTR region of LDLR gene and a TTA insertion 3بIntroduction: Coronary artery disease (CAD) is one of the leading causes of death and disability worldwide. Individual susceptibility to the development of CAD is determined by the interaction between genetic and environmental factors. Therefore the purpose of this study was to investigate the association between the risk of Coronary Artery Disease and LDLR, PCSK9 and SMARCA4 gene polymorphisms and also, severity of stenosis (Number of vessels involved) in patients with coronary artery disease. Material and methods: In this case - control study, 332 subjects included 202 CAD patients and 130 controls were studied. LDL receptor, PCSK9 promoter and SMARCA4 genes polymorphisms were analyzed by PCR and Direct DNA sequencing. Results: This study showed that rs1799898, rs6413504, rs2738464, rs3745678 and rs7259278 in LDLR gene and rs1529729, rs4300767, rs10411252 andrs1122608 in SMARCA4 gene and also, rs2495497 in PCSK9 gene were significantly associated with the risk of CAD (P<0.05). In addition, four polymorphisms in LDLR gene and one inSMARCA4 gene were significantly associated with severity of CAD (P<0.05). In this study, some new variations have been identified which have been included three transition changes; G>A in exon 11, C>T in exon 16 and C>T in 3
