مقدمه و بیان مسئله: مصرف مت آمفتامین با تاثیر بر روی سیستم عصبی مرکزی منجر به بالا رفتن سطح دوپامین در مغز شده و باعث تحریک سلول¬های مغزی می¬شود و در نتیجه منجر به افزایش حالت تهاجمی و حرکات جسمی می¬شود. به خاطر شدت درد التهابی ناشی از ترك، عموما ترك اعتیاد سخت است. امروزه یكی از داروهای پرکاربرد در کاهش درد التهابی ناشی از اعتیاد، بوپره نورفین است. این ماده برای درمان وابستگی به اپیوئید ها نتیجه اثر بخشی دارد. TLR1 و TLR2 گروهی از پروتئین¬ها هستند که نقش مهمی در التهاب و درد التهابی دارند. دراین مطالعه کوشیده¬ایم تا میزان تغییرات بیان ژن این دو عامل را در رت¬های نر معتاد به مت آمفتامین و تحت درمان با بوپره¬نورفین بررسی نماییم. برای القای درد التهابی میزان مناسبی از فرمالین به پنجه¬ی پای جانور تزریق گردید. پس از اتمام آزمایشات، بافت ساقه¬ی مغز از جانوران استخراج شد و میزان بیان دو ژن TLR1 و TLR2 با استفاده از تكنیک Real- time PCR در بافت ساقه¬ی مغز رت¬های نر معتاد به کریستال¬مت و تحت درمان با بوپره¬نورفین بررسی شد.مواد و روش¬ها: 84 سر رت نر نژاد ویستار با محدوده¬ی وزنی (50±200) گرم به صورت تصادفی در 12 گروه 7 تایی شامل گروه کنترل، گروه شاهد، گروه کریستال با دوزmg/kg 10 به مدت 5روز، گروه بوپره نورفین با دوز mg/kg 6 به مدت 5 روز، گروه بوپره نورفین با دوز mg/kg 10 به مدت 5 روز، گروه بوپره نورفین با دوزmg/kg 6 به مدت 14 روز، گروه بوپره نورفین با دوز mg/kg 10 به مدت 14 روز، گروه کریستال با دوزmg/kg 10 و بوپره¬نورفین با دوز mg/kg 6 به مدت 5 روز، گروه کریستال با دوز mg/kg 10 و بوپره نورفین با دوز mg/kg 10 به مدت 5 روز، گروه کریستال با دوز mg/kg 10 و بوپره نورفین با دوز mg/kg 6 به مدت 14 روز، گروه کریستال با دوز mg/kg 10 و سندرم محرومیت خود به خودی به مدت 72 ساعت تقسیم شدند. روش تزریقات درون صفاقی بود . از ساقه مغز تمامی حیوانات نمونه برداری به عمل آمد. سپس میزان بیان دو ژن TLR1 و TLR2 با استفاده از تكنیک Real- time PCR سنجش گردید. به منظور آنالیز داده ها از آزمون One-way ANOVA و نرم افزار SPSS نسخه 22 استفاده شد.یافته¬ها: بر اساس نتایج به دست آمده از آنالیز داده¬های حاصل از واکنش Real time PCR ، نشان داده شد که بیان ژن TLR1 در بافت ساقه مغز در گروه معتاد به مت آمفتامین با دوز 10mg/kg به مدت 5روز متوالی اختلاف معنی داری با گروه کنترل وجود داشت و مقدار بیان آن افزایش یافت (p ‹0.05) ولی بیان TLR2 در این گروه تغییری نداشت. همچنین اختلاف معنی¬داری بین بیان ژن TLR1 در گروه معتاد به مت¬آمفتامین با دوز mg/kg 10 به مدت 5 روز متوالی و سندرم محرومیت خود به خودی با گروه کنترل وجود داشت و مقدار بیان ژن مذکور کاهش یافت ولی اختلاف معنی دار بین این گروه برای ژن TLR2 وجود نداشت.(P›0.05)علاوه بر این نتایج به دست آمده از آنالیز داده¬های حاصل از واکنش Real time PCR ، نشان داد که تزریق بوپره نورفین با دوزهای (6mg/kg و10mg/kg ) که به صورت حاد و مزمن در روزهای مختلف (5 روزه و14 روزه) استفاده شده بود به جز کاهش در مقدار بیان ژن TLR2 در گروه دریافت کننده دوز حاد بوپره نورفین 6mg/kg تاثیر معنی¬داری در بیان ژن TLR1 و TLR2 در ساقه مغز ایجاد نكرد.در پایان نامه حاضر بر اساس نتایج به دست آمده از آنالیز داده¬های حاصل از واکنش Real time PCR، نشان داده شد در گروه های معتاد به مت آمفتامین با دوز 10mg/kg و درمان با بوپره¬نورفین با دوز 6mg/kg به مدت5 روز متوالی، معتاد به مت آمفتامین با دوز 10mg/kg و درمان با بوپره نورفین با دوز 10mg/kg به مدت 5 روز ، معتاد به مت آمفتامین با دوز 10mg/kg و درمان با بوپره نورفین با دوز 6mg/kg به مدت 14 روز نسبت به گروه کنترل اختلاف معنی¬داری در بیان ژن TLR1 وجود ندارد. ولی در گروه معتاد به مت آمفتامین با دوز mg/kg10 و درمان با بوپره نورفین با دوز 10mg/kg به مدت 14 روز نسبت به گروه کنترل اختلاف معنی¬داری در بیان ژن TLR1 وجود دارد و مقدار آن افزایش می¬یابد. (p ‹0.05)علاوه بر این نتایج آنالیز داده¬های حاصل نشان داد که در گروه معتاد به مت آمفتامین با دوز 10mg/kg و درمان با بوپره نورفین با دوز 6mg/kg به مدت 14 روز همانند گروه معتاد به مت آمفتامین با دوز mg/kg10 و درمان با بوپره نورفین با دوز 10mg/kg به مدت 14 روز نسبت به گروه کنترل اختلاف معنی¬داری در بیان ژن TLR2 وجود دارد و مقدار آن¬ها کاهش می¬یابد.بحث: طبق نتایج بدست آمده در این پژوهش، تزریق درون صفاقی مت¬¬آمفتامین به صورت مزمن و دوره محرومیت از مت آمفتامین انجام شد و بیان ژن¬های TLR1 و TLR2 در ساقه مغز تغییرات اندکی نشان داد که به این صورت که بیان TLR1 افزایش و بیان TLR2 کاهش یافت. این حالت بیانگر این است که ممکن است مکانیسم های فیدبکی برای برگشت میزان بیان این ژن به سطوح کنترل وجود داشته باشد. هرچند میزان تغییرات اندک می باشد و نقش قابل توجهی برای این ژن¬ها در ارتباط با اعتیاد به مت¬آمفتامین و درد التهابی درنظر گرفته نمیشود. اما با توجه به حضور این دو ژن در مسیرهای سیگنالینگ درد التهابی و اعتیاد ، ممکن است در قسمت¬هایی دیگر از دستگاه عصبی نقش های واضحتری داشته باشند.

AbstractBackground and aim: Methamphetamine use, by acting on the central nervous system, raises dopamine levels in the brain and stimulates brain cells, resulting in increased aggression and physical activity. It is generally difficult to quit addiction because of the severity of the inflammatory pain caused by quitting.Today, buprenorphine is one of the most widely used drugs in reducing inflammatory pain caused by addiction.It is effective in treating opioid dependence.TLR1 and TLR2 are a group of proteins that play an important role in inflammation and inflammatory pain. In this study, we tried to investigate the extent of gene expression changes in these two factors in male rats addicted to methamphetamine and treated with buprenorphine.An appropriate amount of formalin was injected into the toe of the animal to induce inflammatory pain. However, the rate of variation is small and no significant role for these genes is associated with methamphetamine addiction and inflammatory pain. However, due to the presence of these two genes in the signaling pathways of inflammatory pain and addiction, they may play clearer roles in other parts of the nervous system.
