A general consensus on infrafamiliar classification of Scrophulariaceae is that lacking of great variation of morphological characters in the family did not allow an anambiguous choice of suitable characters for classification. However, anatomy, embryology, cytology and chemistry have been recently more and more introduced as new disciplines in the taxonomy of this family. In particular, owning to the rapid development of the knowledge of chemical constituents, chemosystematics have been proposed as an useful tool providing important additional characters for the classification of members of Scrophulariaceae. The genus Scrophularia of the family Scrophulariaceae comprises about 300 species of herbaceous plants commonly known as figworts. The genus is found throughout the Northern Hemisphere, but concentrated in Southeast of Asia with only a few species in Europe and North America. Characteristic for the genus Scrophularia is the accumulation of iridoid glycosides like aucubin, catalpol, and harpagoside. In addition, many phenylpropanoid glycosides and phenylethanoid glycoside have been isolated from members of the genus Scrophularia. The dried roots of Scrophularia species have been used in Oriental medicine as a treatment for fever, swelling, constipation, pharyngitis, neuritis, and laryngitis. The aim of the present study was the phytochemical investigation of Scrophularia azerbaijanica and Scrophularia variegata ssp. variegata that are indigenous to East Azarbaijan province. In order to purify of secondary metabolites, aerial parts and roots of plants, were soxhlet extracted, with n-hexan, dichloromethane and methanol, successively. The methanolic extract was purified by HPLC apparatus using ODS column. The structure of the purified compounds were determined by spectroscopic method (1HNMR) and confirmed by comparison with reference data. The study of the phytochemical of Scrophularia azerbaijanica led to the isolation of five major compounds, including two iridoids, 6-[4''-O (p-MeO)-E-cinnamoyl]-rhamnopyranosyl catalpol and [6-O-(2",3"-diacetyl,4"-E-cinnamoyl)-rhamnopyranosyl]-catalpol together with three phenylethanoid glycosides, verbascoside (acteoside), methyl verbascoside and martynoside. Furthermore, one iridoid glycoside, [6-O-(2"-vanilloyl)-rhamnopyranosyl] catalpol, and one phenylethanoid glycosides, martynoside, were isolated from Scrophularia variegata. Such identification of the iridoids and phenylethanoid glycosides in two species of Scrophularia make the supplementary search for their physiological roles and taxonomic applications necessary.
